Abberior Expert Line easy3D STED is a microscope with four excitation lasers (405, 485, 561, 640 nm), two channels for spectral detection based on APD technology, and a pulsed STED laser at 775 nm. Included are: adaptive optics for aberration correction, adaptive illumination modes, continuous hardware autofocus for STED and confocal mode, an incubation chamber for live cell imaging, oil and water objectives.
The device was purchased with the funds of the EU project Investing in science and innovation - "IPSted - Innovative microscopy protocols for interdisciplinary research in biomedicine", KK.01.1.1.04.0057.

• STED base: Olympus IX83 inverted microscope with widefield LED illumination (405 nm, 485 nm, 561 nm, 640 nm) and a widefield monochrome camera (1/2" CCD, 1280x960 px)
• Stage with ultrasonic XY scanning and Z-Piezo P-736 scanner with piezo motors with a resolution of 0.7 nm and a range of 200 μm. Z-drift compensation for confocal and STED mode (auto-focus)
• Objectives: (1) 60× / NA 1.2 water, Olympus UPLSAPO 60XW, 0.28 mm working distance; (2) 100× / NA 1.4 oil, Olympus UPLSAPO 100XO 0.13 mm working distance. Correction for coverslip: 0.17 mm.
• Excitation lasers: (1) 405 nm (cw, 50 mW); (2) 485 nm (pulsed, ~ 1 mW at 40 MHz); (3) 561 nm (pulsed, ~ 200 μW at 40 MHz pulse length ~120 ps); (4) 640 nm (pulsed, ~ 1 mW at 40 MHz)
• Spectral detection module: two-channel spectral detection consisting of 2 high-precision single-photon APD detectors with the possibility of time gated detection; tunable spectral bands between 400 and 800 nm; up to 65% photon detection efficiency and low number of dark ones (< 250 Hz); motorised pinhole adjustment (16 openings from 25 μm to 2 mm)
• STED optical package: complete self-adjustment of all beams and pinholes. STED laser 775 nm (pulsed, 2750 mW at 40 MHz; repetition rate 25-40 MHz); STED resolution < 100 × 100 × 100 nm for 3D-STED and < 30 × 30 nm for 2D-STED (typically 80 × 80 × 90 nm and 25 × 25 nm)
• SLM (spatial light modulator) for dynamic beam formation (phase and aberration control)
